I didn't read all of the responses, so this may have been covered, but the only correct answer to the question of what's the most important safety rule is #1 in TKOL's response: Every gun is always loaded (or at least you should treat every gun as if it is loaded). All of the other rules are subsidiary to this one, and if this rule were followed religiously by everyone handling a gun, then the rest of the rules should take care of themselves.

very similar to my treat never keep keep rule mentioned earlier. im just gonna say that most people should be able to handle more than one rule...if not dont own a weapon...but im sure most people on here at least can. if you havnt already...learn one of these little ditties and you'll be just fine.

Didn't read all the posts but, by far this is the most important safety, the gun won't fire if your fingers off the trigger. Never mind any arguments about dropping and it going off, and the like, those are issues of gun quality, and being a douche bag for letting your weapon fall from your hand. Of course this is all IMHO.

Common sense says, never point a gun at someone your not intending to shoot.
